Case Transgenic and Targeting Core

The Case Transgenic and Targeting Facility provides transgenic, targeting (knockouts, knockins, targe ted point mutations, conditional knockouts), chimera, rederivation and cryopreservation services for the Cleveland Biomedical Research Community. The Facility Director is Ronald Conlon of the Department of Genetics.

The Case Transgenic and Targeting Facility office is located in Wolstein Research Bldg 2521.

Services are contracted on a fee-for-service, first-come-first-serve basis with an oustanding record of success.

Bioinformatics Core

A bioinformatics core facility is located one the first floor of the Wolstein Research Building. The current system configuration consists of 20 dual processor Apple XServe G5's, an 8 Terabyte capacity File server, a dedicated database server, and a portal system for easy centralized access. Each compute node is outfitted with two GHz G5 processors and four Gb of RAM. The PC cluster farm has sufficient disk and computational capacity to manipulate sequence of entire genomes and to perform complex phylogenetic and comparative sequence analyses. The cluster provides the backbone of the computational support of the Department of Genetics at Case Western Reserve University.

Sequencing support for software (Miropeats, EMBOSS, Consed, FASTA, NCBI blast, and others) has been installed on the core computers and is used for the analysis, assembly and storage of large-scale sequencing data. The computational facility is directly connected to the university's campus-wide gigabit network. In addition, the cluster operates within its own private gigabit ethernet backbone to facilitate efficient intranet exchange of data. Finally, the entire system is accessible via a web interface allowing users easy point and click access to the tools installed.

The Genetics facility runs alongside the Case Bioinformatics Core Facility. The Case Bioinformatics Core Facility is dedicated to support the continuing research and education mission of the School of Medicine and the Comprehensive Cancer Center at CASE. It is aimed at supporting the medical, biomedical and research scientist, residents, fellows and graduate students in their research projects in the specific fields of Biostatistics, Bioinformatics, and Data Mining, as applied to medical, biomedical, biological and environmental projects. Services to investigators come in the form of consultation or collaboration. In a typical year we expect to provide more than 1,500 consultant-hours of service.

Imaging Core

The Imaging Facility is a light microscope facility dedicated to bringing state-of-the-art methods in modern imaging to our biomedical and research scientists here at CASE. The Imaging Facility consists of nine complete microscope workstations (6 widefield, 2 of which are completely motorized and automated; 1 widefield equipped for live-cell imaging and deconvolution; 1 confocal equipped for live-cell imaging; and 1 fluorescence stereomicroscope). All of the widefield microscopes are equipped with CCD cameras, two of which are color cameras to aid in histological analysis. Lenses are from 1.5x through 100x, with optics for Brightfield, Darkfield, Phase, Polarization, DIC, Hoffman Modulation Contrast, and Fluorescence. The lasers on the confocal are 405 nm, Argon (458, 476, 488, 496, 514 nm), Green HeNe (543 nm), Orange HeNe (594 nm), and Red HeNe (633 nm). There is also a remote workstation for analysis. Software processing programs include those for visualization (2-D and 3-D), publication, measurement and tracking, and deconvolution.

Shared Instrumentation Grants from NIH NCRR (numbers RR-017980 and RR-021228) allowed us to purchase our Leica TCS SP2 AOBS filter free UV/spectral confocal laser scanning microscope and the two Leica widefield automated and motorized microscopes. Departmental funds allowed us to purchase the Leica multi-dimensional workstation for live-cell imaging. HHMI provided the funds for the GFP stereomicroscope and two of the widefield setups.

DNA sequencing Core

Located on the 7th floor of the BRB, the Genomics Core Facility provides sequencing and genotyping capabilities that is available to all investigators at Case. Custom designed SNP projects as well as human and mouse linkage analysis projects can also be handled by the Core.

The Core is equipped with an ABI Prism 3730 DNA Analyzer which handles the majority of all sequencing samples and two ABI Prism 3100 Analyzers for genotyping samples. The Illumina Beadstation 500GX Genetic Analysis System runs all custom designed SNP projects and linkage projects using BeadArray technology on two different Array of ArraysTM formats, the Sentrix® Array Matrix and the Sentrix® BeadChip. Large-scale genome-wide SNP association studies are performed using Illumina\'s Infinium assay.

The mission of the Core is to provide better data at a reduced cost and turn-around time with respect to outside sources.
